Francisco de Madrid came to New Mexico in 1603, and brought 10 new soldiers and four friars along with him. In 1626 he boasted that he was one of the "ancient settlers." At this time he is mentioned as the brother-in-law of Pedro Márquez. His wife was María de la Vega Márquez.

By 1639, he was a Captain when he gave his age as forty-six. In 1641, Francisco was a member of the Santa Fé Cabildo, or town council.

His children to all appearances, were: Francisco II, Francisca, wife of Juan Varela de Losada; and María, nicknamed "Mariaca." An Alférez, Cristóbal de Madrid, who escorted some colonists to New Mexico in 1641, might have been another son, or perhaps the elder Francisco was meant since Cristóbal is not heard of again.

Francisco married María de La Vega Márquez, daughter of Gerónimo Márquez and Unknown, about 1593. (María de La Vega Márquez was born in 1597 and died about 1660.)
