Isabel was one of three daughters of John de Bolton. Isabel's sisters were Catherine, who married Simon de Pudsey, and Christian who married William de Rimington. John de Bolton gave Isabel lands in Midhope, and on 24 Sep 1329, "John fil. John de Boulton" releases th "Isabella filia mee" all title to certain lands there.

"Isabel de Bolton, it is believed was descended through the illustrious families of Clare, Gant, and roumere from the old Saxon Earls of Mercia, William Roumere, one of the great Norman Barons, having after the Conquest, married Lucy, sister and heir of Edwin, the last Earl, and thereby possessing, as it is quoted by Sir William Dugdale, 'Cravennam, et Couplendiam, et Allerdale, et Cocerham jur hereditario.' [Dugdale, Monast., i. p. 798]

According to the author of ~Memorials of an Ancient House : a History of the Family of Lister or Lyster,, p. 4, the Heröologic Listeriensis gives her descent as:

In 50 Edward III (1376), Isabel, daughter of John de Bolton, and once wife of Henry de Cliderow, grants to her son, Richard Lyster twelve messuages in Middope, and, 31 Oct 1376, she releases to this Ricahrd all the lands she had of feoffment of her father in Middope. She had married, as first husband, Henry de Cliderow, who died before Oct 1376, and had issue by him of Edmund, the father of Nicholas de Cliderow, who, 28 Oct 1379, released to Richard Lyster the lands, in Middope which he had of John de Bolton.
